Study on the growth-promoting properties of tilapia by Eucommia extracts

Separately using Eucommia extracts, Colistin and Bacitracin Znic with diet to do feeding contrast test for 540 tilapias. Colistin and Bacitracin Znic are common antibiotic feed additives. The test is from average initial weight 45g growing to 1500g during 150 days. The results show that using 1500mg/kg Eucommia extracts as additives to feed tilapias, the relative growing speed is 2.16% higher than Bacitracin Znic, but the differences aren't significant(P>0.05). The relative growing speed is 29.42% and 19.60%, Blank control and Colistin and it has significant differences(P<0.05). Bait Coefficient: The test groups of Eucommia extracts which add in 1000mg/kg and 1500mg/kg have not significant differences with the Bacitracin Znic group(P>0.05), but have significant differences with Blank Control group (P<0.05).  Survival rate: The survival rate of feeding group with Eucommia extracts and Colistin as additive is 100%. The blank group and Bacitracin Znic group is 98.89%. but through variance analysis, there haven't significant differences among the groups(P>0.05).
The blood cholesterol in Tilapia Mozambicans which were fed with Eucommia extracts is lower than the Blank control, and the group fed with the additive of Colistin and Antibiotics. It is lower 53.95% than the Blank control, 37.06% and 5.48% than that feed with Bacitracin Zinc and Colistin. The difference is significant. The serum globulins of that Tilapia Mozambicans fed with Eucommia extracts is higher than the Blank control and feed with antibiotics. It is higher 17.14%, 24.58% and 8.97% than the Blank control, Colistin and Bacitracin Zinc, and the difference is not significant.

2  The function of against common fish pathogens of Eucommia extracts
The experiment shows that Eucommia extracts(0.2g/ml, 8g active ingredient) can effectively inhibit the growth of the Aeromonas punctta f. intestinalis and Myxococcs strains 58-20-9,56-12-10  and binary fusants and ternary fusants . This concentration Eucommia extracts also can kill the living 56-12-10 .From all of that, it can be ensured that Eucommia extracts have resistance to common fish disease of bacterial gillrot disease, bacterial enteritis, red skin disease.

3  Eucommia extract's mould proof effect study
Add 0.15% pure Eucommia powder (as active ingredient 8g) to feed, it can effectively prevent the feed mildewing. The mold multiply in feed, which kept for 45~60days in the condition: between July and September, at room temperature 27-38°(average 31.5°), relative humidity 50-84%.Without preservatives feed, the total mold is 5.0 × 10⁵cfu/g. added pure Eucommia powder feed, the total mold is 1.7 x 10⁵cfu/g. But the feed with Luprosil Salt (main calcium propionate) and Confeed (composite mold inhibitor), the total mold is 2.3×10⁵cfu/g and 2.8 × 10⁵cfu/g. The result indicate that feed with pure Eucommia powder, its total mold is significantly lower than the other three groups (P <0.01), the inhibition rate of mold by pure Eucommia powder is 78.2%.

4 Safety evaluation test report
Toxicological safety determination of pure Eucommia powder
1. General toxicological test
Acute toxicity test
Do acute toxicity test on Kunming mice, which are provided by Hunan Institute of Pharmaceutical Industry. With high concentration solution of pure Eucommia powder to irrigation stomach, determinate its LD50. The result turn out that there is no death and could not determinate LD50. The oral maximum tolerated dose (MTD) is greater than 27788.0mg/kg, weight (1.54-1.74 times mouse body weight) LD50 of intraperitoneal injection is 2610.0mg/kg. It indicates that the pure Eucommia powder is very low toxicity.

The chronic toxicity tests of Eucommia bark extract (pure powder)
According to the maximum tolerance dose of acute toxicity test, use 6.17, 2.06, 0.69g/kg Eucommia bark extract on mouse respectively, take for 8-week, results showed that it has no effects on mouse and its Blood conventional index; and there is no significant difference on normal blood with the control group (P > 0.05), but Eucommia bark extract has certain inhibition of growth on mouse if it takes high dose, the content of total protein, albumin, urea nitrogen in the blood was significantly higher than other groups, however, low doses of eucommia bark extract on the growth of the middle mouse has a significant role in promoting, the activity  of GOT, GPT and ALP in the blood will decline with the increase of the dose, and there has very significant difference with blank control group(P < 0.01). So, eucommia bark extract  can reduce the blood of the active, improves the digestive system function.  Low doses of eucommia bark extract can reduce the content of urea nitrogen in the blood, improve the efficiency of nitrogen absorb, and no adverse effects to other organs.
